چکیده

The levels of leukocyte adenine phosphoribosyltransferase and of hypoxanthine-guanine phosphoribosyltransferase (H-GPRT) were compared in normal control subjects, patients with acute leukemia in remission, and patients with acute leukemia in exacerbation. Significant (p < 0.025) elevations of both the adenine phosphoribsosyltransferase and H-GPRT activities occurred in individuals with leukemia in relapse, compared with those in remission. Altered ratios of the specific enzymatic activities of adenine phosphoribosyltrans ferase to H-GPRT were noted in two of eight 6-thiopurineresistant patients. Leukemic cells from one patient with chronic myelogenous leukemia in blast crisis exhibited a marked decrease in H-GPRT activity. The H-GPRT from the second patient showed an altered affinity for purine bases and, most markedly, for 5'-phosphoribosyl-1-pyrophosphate when

Inhibition by 6-mercaptopurine of Purine Phosphoribosyltransferases from Ehrlich Ascites-tumour Cells That Are Resistant to the Drug.

1. A strain of Ehrlich ascites-tumour cells that showed little inhibition of growth in the presence of 6-mercaptopurine accumulated less than 5% as much 6-thioinosine 5'-phosphate in vivo, in the presence of 6-mercaptopurine, as did the sensitive strain from which it was derived. 2. Specific activities of the phosphoribosyltransferases that convert adenine, guanine, hypoxanthine and 6-mercaptop...

The substrate specificity of purine phosphoribosyltransferases in Schizosaccharomyces pombe.

1. The activities of the purine phosphoribosyltransferases (EC 2.4.2.7 and 2.4.2.8) in purine-analogue-resistant mutants of Schizosaccharomyces pombe were checked. An 8-azathioxanthine-resistant mutant lacked hypoxanthine phosphoribosyltransferase, xanthine phosphoribosyltransferase and guanine phosphoribosyltransferase activities (EC 2.4.2.8) and appeared to carry a single mutation. Two 2,6-di...

Photolysis of Thiopurines in the Presence of Oxygen

Thiopurines, Photolysis, Photooxygenation, Purine Derivatives Irradiation (A > 290 nm) of 6-thiopurine and some 9-substituted 6-thiopurines in oxygen saturated iertf-butanol leads to hypoxanthine, purine and purine-6-sulfonic acid or their substituted derivatives. Purine and hypoxanthine formation can be sensitized with dyes. The reaction of 6-thiopurine with singlet oxygen has also been perfor...

Structure-activity relationships and cross-resistance observed on evaluation of a series of purine analogs against experimental neoplasms.

Extensive efforts by the Burroughs-Wellcome group, the Sloan-Kettering group, and others have led to a considerable knowledge of purine antagonists that inhibit the growth of bacteria, experimental neoplasms, and human leukemias (18). Although 6-mercaptopurine (6-MP) is the most widely employed purine antagonist from the clinical standpoint, "related compounds, thioguanine, 6-chloropurine, 6-me...
